Background
SEZ6 (Seizure-related gene 6) is a type I transmembrane glycoprotein (~110 kDa) composed of five short consensus repeat (SCR) domains, three CUB domains, a single transmembrane segment and a short cytoplasmic tail that is predominantly expressed in the central nervous system, where it acts as a critical trafficking chaperone for kainate receptor subunits GluK2/3, ensuring their proper N-glycosylation (including the HNK-1 epitope), post-ER export and surface localization to regulate synaptic connectivity, long-term potentiation and dendritic arbor balance, and is sequentially cleaved by BACE1 and γ-secretase to release a soluble ectodomain whose CSF level serves as a direct pharmacodynamic read-out of BACE1 inhibition, while in neuroendocrine neoplasms its selective over-expression is exploited as an ADC target for therapies against small-cell lung cancer and related tumors.

IHC shows positive staining in paraffin-embedded human cerebral cortex. Anti-SEZ6 antibody was used at 1/2000 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.
