Background
SERPINB3, also known as Squamous Cell Carcinoma Antigen 1 (SCCA1), is a member of the clade B serpin family that functions primarily as an intracellular inhibitor of cysteine and serine proteases, playing a critical role in regulating apoptosis and protecting cells from protease-induced damage. Normally expressed in stratified squamous epithelia such as the skin, cervix, and lung, this protein helps maintain tissue integrity by inhibiting enzymes like cathepsins and papain-like proteases; however, its expression is frequently and significantly upregulated in various malignancies, particularly squamous cell carcinomas of the head and neck, lung, esophagus, and cervix, where it contributes to tumor progression by conferring resistance to chemotherapy and radiation through the suppression of granzyme B-mediated cytotoxicity and the inhibition of Fas-mediated apoptotic pathways. Due to its strong association with advanced disease stages, poor prognosis, and therapeutic resistance, SERPINB3 serves as a valuable clinical biomarker for monitoring tumor burden and recurrence, while also emerging as a potential therapeutic target aimed at restoring apoptotic sensitivity in resistant cancer cells.

IHC shows positive staining in paraffin-embedded human esophagus. Anti-SERPINB3 antibody was used at 1/2000 dilution, followed by a HRP Polymer for Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.
