Background

Sec61B is a critical subunit of the Sec61 translocon complex, which serves as the primary protein-conducting channel in the endoplasmic reticulum (ER) membrane of eukaryotic cells. As part of the heterotrimeric Sec61 complexcomposed of Sec61α, Sec61β (Sec61B), and Sec61γ—Sec61B plays an essential structural and regulatory role in facilitating the co-translational translocation of nascent polypeptide chains into the ER lumen or their integration into the ER membrane. Although it is the smallest subunit, Sec61B stabilizes the complex by interacting with Sec61α and helps maintain the integrity of the translocation pore, thereby ensuring efficient protein secretion and membrane protein biogenesis. Beyond its canonical role in protein translocation, emerging evidence suggests that Sec61B may also participate in ER stress responses, calcium homeostasis, and the regulation of channel gating, highlighting its multifaceted importance in cellular proteostasis and overall ER function.

Immunohistochemistry

IHC shows positive staining in paraffin-embedded human kidney. Anti-Sec61B antibody was used at 1/1000 dilution, followed by a HRP Polymer for Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.
