Background

SEC24D (SEC24 homolog D) is a key cargo-selection subunit of the COPII vesicle coat, encoded by the SEC24D gene located on chromosome 4q26, with a protein of 1,032 amino acids and a molecular weight of approximately 115 kDa. Structurally, SEC24D contains a conserved zinc finger domain, a Sec23/Sec24 trunk domain, a β-sandwich domain, a helical domain, and a gelsolin-like domain, forming multiple cargo-binding sites including an IxM-binding pocket. Its core function is to form a heterodimer with SEC23 at endoplasmic reticulum exit sites, selectively packaging transmembrane cargo proteins into COPII vesicles by recognizing specific motifs in their cytoplasmic tails, thereby mediating anterograde transport of secretory proteins from the endoplasmic reticulum to the Golgi apparatus. SEC24D exhibits unique cargo selectivity: its IxM-binding pocket can recognize cargo such as the SNARE proteins GOSR2 and STX5 that contain IxM motifs, and it can also specifically package GPI-anchored proteins through the cargo receptor TMED10. Clinically, mutations in SEC24D are closely associated with Cole-Carpenter syndrome type 2, an autosomal recessive bone formation disorder characterized by craniofacial malformations, defective skull ossification, and susceptibility to fractures. In addition, SEC24D has been identified as a susceptibility gene for hepatitis B virus infection and shows aberrant expression and functional necessity in various tumors.
