Background
SEC24C (SEC24 homolog C, component of the COPII coat complex) is a key cargo-selection subunit of the COPII vesicle coat, encoded by the SEC24C gene located on chromosome 10q22.2, with a protein of 1,125 amino acids and a molecular weight of approximately 118 kDa. Structurally, SEC24C contains conserved zinc finger, β-sandwich, helical domains, and a gelsolin-like domain, forming multiple cargo-binding pockets. Its core function is to form a heterodimer with SEC23 at endoplasmic reticulum exit sites, selectively recognizing transmembrane cargo proteins through di-acidic or di-hydrophobic motifs in their cytoplasmic tails, concentrating them into newly formed COPII vesicles, and thereby mediating anterograde transport of secretory proteins from the endoplasmic reticulum to the Golgi apparatus. SEC24C exhibits unique cargo selectivity: its surface groove can bind IxM motifs (such as in the SNARE proteins GOSR2 and STX5), and it specifically packages GPI-anchored proteins through the cargo receptor TMED10. In addition, SEC24C is involved in HIV-1 viral replication as an important host dependency factor, and in neurons it maintains proteostasis and polarity by transporting ion channels and receptors.
