Background
SCP3, also known as Synaptonemal Complex Protein 3, is a meiosis-specific protein encoded by the SYCP3 gene. As a major constituent of the transverse filaments of the synaptonemal complex, it plays a core structural and functional role in homologous chromosome pairing, synapsis, and recombination during meiotic prophase. The protein assembles into homotetramers that align head-to-tail to form fiber structures with approximately 20 nm transverse striations, and interacts with proteins such as SYCP2 to maintain the integrity of the lateral elements of the synaptonemal complex. Its N-terminal Cor1 domain and coiled-coil domain are critical for mediating protein homotypic interactions and DNA binding. In terms of physiological function, SCP3 is essential for male gametogenesis; its deficiency leads to spermatogenic arrest at the pachytene stage, resulting in azoospermia. In females, although its function is not absolutely essential, its absence also affects fertility, leading to chromosomal aneuploidy and embryonic lethality. At the clinical-pathological level, SYCP3 gene mutations have been confirmed as a genetic factor in male spermatogenic failure (SPGF4) and female recurrent pregnancy loss. Moreover, in various cancers—particularly cervical cancer—aberrant overexpression of SCP3 significantly promotes tumor progression through activation of the AKT signaling pathway, and is closely associated with poor prognosis and shorter overall survival in patients, making it a potential therapeutic target for cancer treatment.
