Background

SCD (stearoyl-CoA desaturase) is an integral membrane protein localized to the endoplasmic reticulum membrane and is the rate-limiting enzyme in the biosynthesis of monounsaturated fatty acids (MUFAs), catalyzing the introduction of the first cis double bond at the Δ9 position of saturated fatty acyl-CoAs. Humans possess two homologous proteins, SCD1 and SCD5, whereas mice express four isoforms, SCD1 through SCD4. The enzyme uses palmitoyl-CoA and stearoyl-CoA as its principal substrates, converting them to palmitoleoyl-CoA (16:1) and oleoyl-CoA (18:1), respectively, and these monounsaturated fatty acids are key precursors for the synthesis of membrane phospholipids, cholesteryl esters, and triglycerides. The catalytic mechanism of SCD depends on a di-iron center coordinated by nine conserved histidine residues, which activates molecular oxygen, and it also requires cytochrome b5 as an electron donor to regenerate the iron center. Structural biology studies have revealed that SCD1 adopts a unique fold: four transmembrane helices form a conical structure capped by a cytosolic domain, with the substrate acyl chain enclosed within a narrow tunnel approximately 24 Å long that contains a sharp bend at the C9 and C10 positions, thereby precisely determining the positional selectivity and stereospecificity of the desaturation reaction. Functionally, SCD1 knockout mice exhibit a lean, hypermetabolic, insulin-sensitive phenotype with resistance to diet-induced obesity, making SCD an attractive drug target for metabolic diseases such as obesity, diabetes, and non-alcoholic fatty liver disease. At the same time, SCD is overexpressed in a variety of malignant tumors, supporting tumor growth by supplying monounsaturated fatty acids required for membrane biosynthesis and by participating in the lipidation of Wnt signaling pathway ligands, making it a highly watched target in the field of cancer therapy as well.
