Background
SCAMP1 (Secretory Carrier Membrane Protein 1) is a ubiquitously expressed, four-pass transmembrane protein that cycles between the trans-Golgi network, endosomes and the plasma membrane, where it acts as a multifunctional regulator of post-Golgi membrane trafficking, exocytosis and cell-surface recycling; its cytosolic N- and C-terminal domains contain NPF motifs, a proline-rich SH3-binding site and three conserved amphiphilic segments that dock at the cytoplasmic membrane interface, enabling SCAMP1 to nucleate protein–protein interactions with SNAREs and other trafficking factors, thereby facilitating secretory granule exocytosis in neuroendocrine cells and insulin-secreting β-cells, while in epithelia it cooperates with the metastasis suppressor MTSS1 to stabilize Rac1-based cell–cell adhesions and curb cancer invasion, and palmitoylation of its cytosolic segments dynamically modulates these membrane exchange and anti-invasive functions.
