Background

SAMHD1 (Sterile Alpha Motif and HD Domain-Containing Protein 1) is a protein that plays a crucial role in cellular processes. It is involved in the regulation of dNTP (deoxyribonucleoside triphosphate) levels within cells, which is essential for DNA synthesis and repair. SAMHD1 acts as a dNTP triphosphohydrolase, helping to maintain a balanced pool of dNTPs to prevent excessive DNA replication and genomic instability. Additionally, it has been implicated in innate immune responses, particularly in restricting the replication of certain viruses, such as HIV-1, by limiting the availability of dNTPs required for viral reverse transcription. Its dysfunction has been associated with various diseases, including Aicardi-Goutières syndrome, an autoimmune disorder characterized by chronic inflammation and neurological damage.
