Background

There are two types of cytoskeletal and microfibrillar keratin: I (acidic; 40-55 kDa) and II (neutral to basic; 56-70 kDa).Keratin 10 plays a role in the establishment of the epidermal barrier on plantar skin.

IP

Keratin 10 Rabbit mAb at 1/25 dilution (0.5 µg) immunoprecipitating Keratin 10 in 0.2 mg K562 whole cell lysate.
Western blot was performed on the immunoprecipitate using Keratin 10 Rabbit mAb at 1/1000 dilution.
Secondary antibody (HRP) for IP was used at 1/400 dilution.

Immunohistochemistry

IHC shows positive staining in paraffin-embedded human skin. Anti-Keratin 10 antibody was used at 1/1000 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.
