Background

Ribosomal Protein S3 (RPS3) is a multifunctional component of the 40S small ribosomal subunit in eukaryotes, primarily recognized for its essential role in protein synthesis by facilitating the accurate decoding of mRNA and maintaining translational fidelity through its interaction with the 18S rRNA. Beyond its canonical function in translation, RPS3 exhibits significant "moonlighting" activities, particularly as an extraribosomal protein involved in DNA repair mechanisms, where it acts as a DNA glycosylase to remove oxidized bases such as 8-oxoguanine, thereby protecting genomic integrity against oxidative stress. Furthermore, RPS3 participates in various cellular signaling pathways, including the regulation of apoptosis and the NF-κB pathway, and has been implicated in viral infection processes, such as serving as a receptor or cofactor for certain viruses, highlighting its critical importance in both fundamental cellular homeostasis and disease pathogenesis.

Immunohistochemistry

Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 4% PFA and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
