Background

Ribosomal Protein S14 (RPS14) is a fundamental component of the small 40S ribosomal subunit in eukaryotic cells, playing a critical role in the structural integrity and functional accuracy of the protein synthesis machinery. Encoded by the RPS14 gene, this highly conserved protein is essential for the proper assembly of the 40S subunit and facilitates key steps in translation initiation and elongation by helping to maintain the correct configuration of the decoding center, thereby ensuring fidelity in mRNA reading and tRNA selection. Beyond its canonical housekeeping function in ribosome biogenesis, RPS14 has gained significant clinical attention due to its location within the commonly deleted region of chromosome 5q (5q-) in myelodysplastic syndromes (MDS); haploinsufficiency of RPS14 in hematopoietic stem cells leads to impaired erythropoiesis and increased apoptosis, directly linking defective ribosome production to the pathophysiology of this specific subtype of bone marrow failure, while also highlighting its emerging roles in extra-ribosomal functions such as the regulation of p53 stability and cellular stress responses.
