Background

RPA3, also known as Replication Protein A3, is the smallest subunit (14 kDa) of the heterotrimeric replication protein A (RPA) complex, encoded by a gene located on human chromosome 7p21.3. It comprises 121 amino acids and features an oligonucleotide/oligosaccharide-binding (OB-fold) domain as its structural core. As the structural scaffold of the RPA complex, RPA3 is essential for the assembly and stability of the entire complex. Through tight association with the RPA1 (70 kDa) and RPA2 (32 kDa) subunits, it forms a single-stranded DNA (ssDNA)-binding platform that plays a central role in nearly all aspects of DNA metabolism, including DNA replication, recombination, and repair. Upon cellular detection of DNA damage, the RPA complex rapidly binds to and stabilizes exposed ssDNA intermediates. This process not only prevents re-annealing of DNA but also activates the ATR-dependent DNA damage checkpoint by recruiting key factors such as the ATRIP protein kinase, and further recruits downstream repair proteins including RAD51 and XPA to damage sites, initiating pathways such as homologous recombination repair and nucleotide excision repair. Additionally, RPA3 can form an alternative RPA complex (aRPA) with RPA4 (30 kDa), potentially functioning in certain specific DNA repair contexts. Aberrations in RPA3 itself or its interacting factors—such as RFWD3-mediated ubiquitination—have also been found to be genetically associated with various diseases including lung cancer, head and neck cancer, and myocardial infarction.
