Background

Receptor tyrosine kinase-like orphan receptor 2 (ROR2) is a transmembrane protein belonging to the ROR subfamily of receptor tyrosine kinases, playing a pivotal role in non-canonical Wnt signaling pathways, particularly the Wnt5a-mediated planar cell polarity and Wnt/Ca2+ pathways. Structurally characterized by an extracellular domain containing cysteine-rich and kringle domains that facilitate ligand binding, a single-pass transmembrane region, and an intracellular tyrosine kinase domain, ROR2 functions as a critical regulator during embryonic development, influencing processes such as skeletal morphogenesis, heart formation, and neural tube closure. Beyond its developmental significance, ROR2 has garnered substantial attention in oncology due to its context-dependent dual role; it can act as a tumor suppressor in certain tissues while promoting metastasis, invasion, and epithelial-mesenchymal transition (EMT) in various cancers, including melanoma, renal cell carcinoma, and osteosarcoma, often by interacting with other receptors like RYK or modulating downstream effectors such as JNK and NF-κB. Furthermore, mutations in the ROR2 gene are directly linked to human genetic disorders, most notably autosomal recessive Robinow syndrome and autosomal dominant brachydactyly type B, highlighting its essential function in limb and facial development, which makes it a promising therapeutic target for both congenital defects and cancer treatment strategies.
