Background

RNA polymerase II (RNAP II and Pol II) is a multiprotein complex that transcribes DNA into precursors of messenger RNA (mRNA) and most small nuclear RNA (snRNA) and microRNA. The purified enzyme has typically 10–12 subunits (12 in humans and yeast). The subunit RPB1 contains a carboxy terminal domain (CTD) composed of up to 52 heptapeptide repeats (YSPTSPS) that are essential for polymerase activity.

Western blot was performed on the immunoprecipitate using RNA polymerase II CTD repeat YSPTSPS (phosphor S7) Rat mAb at 1/1000 dilution.

Immunocytochemistry

ICC shows positive staining in HeLa cells. Anti-RNA polymerase II CTD repeat YSPTSPS (phospho S7) antibody was used at 1/100 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rat IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 100% ice-cold methanol and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI.
