Background

Rheb protein is a small GTPase that plays a crucial role in regulating cell growth and metabolism. It acts as a key activator of the mTOR (mechanistic target of rapamycin) pathway, which is central to controlling protein synthesis, cell size, and autophagy. When Rheb is bound to GTP, it stimulates mTORC1 activity, promoting anabolic processes that support cell growth and proliferation. Conversely, when it is in its GDP-bound state, mTORC1 is inhibited, allowing cells to shift towards catabolic processes. Rheb is involved in various cellular functions and its dysregulation has been implicated in several diseases, including cancer and neurodegenerative disorders.

Immunohistochemistry

IHC shows positive staining in paraffin-embedded human cardiac muscle. Anti-Rheb antibody was used at 1/200 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

IHC shows positive staining in paraffin-embedded human kidney.

IHC shows positive staining in paraffin-embedded human liver.

IHC shows positive staining in paraffin-embedded human cervical squamous cell carcinoma.
