Background

RBM10, also known as RNA-binding motif protein 10, is a multifunctional RNA-binding protein encoded by the RBM10 gene located on the X chromosome, and is widely involved in multiple levels of gene expression regulation, particularly playing a critical role in the alternative splicing of pre-mRNA. This protein contains multiple typical RNA recognition motifs, a C2H2 zinc finger domain, and a nuclear localization signal sequence, which collectively confer its ability to recognize and bind specific RNA sequences, thereby regulating the splicing patterns of target genes. As a splicing regulator, RBM10 typically promotes or inhibits the inclusion of specific exons by binding to its target pre-mRNA, thus modulating the diversity of protein coding sequences; its well-known important targets include genes such as NUMB and FAS, which are associated with cell proliferation, differentiation, and apoptosis. In terms of biological functions, RBM10 is broadly involved in the regulation of embryonic development, cell cycle progression, and apoptosis, and is essential for maintaining normal tissue homeostasis. Notably, mutations in the RBM10 gene are closely associated with TARP syndrome, an X-linked congenital developmental disorder characterized by congenital heart defects, cleft palate, ear anomalies, and developmental delay in affected patients; furthermore, in various tumors (such as lung cancer, colorectal cancer, and glioblastoma), RBM10 frequently undergoes deletion or mutation, and its loss of function promotes tumor progression through aberrant splicing, thus establishing RBM10 as an important tumor suppressor gene. The multifaceted nature of these functions position RBM10 as a key molecule for studying RNA regulatory networks, developmental biology, and cancer pathogenesis.

Immunohistochemistry

IHC shows positive staining in paraffin-embedded human stomach. Anti-RBM10 antibody was used at 1/200 dilution, followed by a HRP Polymer for Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

Immunocytochemistry

ICC shows positive staining in Ramos cells. Anti- RBM10 antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 4% PFA and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
