Background

RBBP4 (Retinoblastoma Binding Protein 4), also known as RbAp48 or NURF55, is a highly conserved core subunit of several essential chromatin-modifying complexes, including the NuRD (Nucleosome Remodeling and Deacetylase) complex, the PRC2 (Polycomb Repressive Complex 2), and the HAT1 histone acetyltransferase complex. Functioning primarily as a histone chaperone, RBBP4 facilitates the assembly and stability of nucleosomes by binding directly to the N-terminal tails of histones H3 and H4, thereby playing a pivotal role in regulating chromatin structure and epigenetic gene expression. Through its involvement in these multiprotein complexes, RBBP4 contributes to critical cellular processes such as transcriptional repression, DNA replication, cell cycle progression, and cellular differentiation, while its dysregulation has been implicated in various pathological conditions, including cancer and neurodegenerative disorders, highlighting its significance as both a fundamental regulator of genome integrity and a potential therapeutic target.
