Background
Rat IgE protein is a ~190 kDa, tetrameric immunoglobulin isotype composed of two identical heavy (ε) chains and two identical light (κ or λ) chains, which, when synthesized by B cells in response to allergens or helminths, is secreted as a cytokine-like mediator that binds with nanomolar affinity to the high-affinity FcεRI receptor on mast cells and basophils, thereby arming them for immediate hypersensitivity reactions; its ε-heavy chain contains four constant-region domains (Cε1–Cε4) that confer the unique ability to interact with FcεRI and CD23 (FcεRII), a hinge-rich Cε2 domain that imparts flexibility, and multiple N-linked glycans critical for folding and half-life, while the variable regions generated through V(D)J recombination confer antigen specificity, making rat IgE an essential experimental tool for modeling human allergic asthma, anaphylaxis, and parasitic immunity, and a key reagent for quantifying allergen-specific antibody responses, studying FcεRI signaling cascades, and evaluating therapeutic candidates targeting the IgE/FcεRI axis.
