Background

Rat IFN-γ (interferon-gamma) is a 15–16 kDa, 136-amino-acid type II cytokine secreted chiefly by activated NK cells and Th1-type CD4⁺ T cells. It signals through a high-affinity heterodimeric receptor composed of IFN-γR1/IFN-γR2, activating the JAK–STAT1 pathway to up-regulate MHC class I and II molecules, chemokines such as CXCL9/10, and multiple antimicrobial effectors. In rats this molecule is encoded by the Ifng gene on chromosome 7 and is heavily N-glycosylated; it functions as a non-covalent antiparallel dimer with species-specific biological activity that is essential for macrophage activation, Th1 polarization, and control of intracellular pathogens.
