Background
Siglec-E, the murine orthologue of human Siglec-9, is a sialic acid-binding immunoglobulin-like lectin expressed predominantly on myeloid cells (macrophages, neutrophils, dendritic cells) that functions as a critical immunosuppressive checkpoint by recruiting SHP-1/2 phosphatases upon engagement of sialoglycan ligands, thereby attenuating TLR4- and TRIF-dependent signaling, dampening NF-κB activation and pro-inflammatory cytokine production, and exerting neuroprotective and anti-atherogenic effects through inhibition of microglial activation and CD36-mediated oxidized LDL uptake.
