Flow cytometric analysis of fresh C57BL/6 mouse bone marrow labeled with anti - Mouse Ly-6B.2 antibody at 1/200 dilution (1 μg) / (right panel) compared with a Rat IgG2a, κ Isotype Control / (left panel). Goat Anti-Rat IgG Alexa Fluor® 488 was used as the secondary antibody. Then cells were stained with CD11b - Alexa Fluor® 647 antibody separately. Flow cytometry and data analysis were performed using Agilent NovoCyte Quanteon and FlowJo™ software.

Background
Ly-6B.2 is an important glycosylphosphatidylinositol (GPI)-anchored protein belonging to the mouse Ly-6 antigen complex family, with a molecular weight of approximately 25–30 kDa, which reduces to about 15 kDa upon removal of N-linked glycosylation. This protein is predominantly highly expressed on the surface of neutrophils, inflammatory monocytes, and partially activated macrophages, and is abundantly present in tissues such as bone marrow, spleen, lung, and lymph nodes. The function of Ly-6B.2 is closely associated with inflammatory responses; its high expression on inflammatory cells makes it an important participant in inflammatory reactions. Studies have shown that Ly-6B-expressing macrophages represent a newly generated population of inflammatory macrophages and serve as a surrogate marker for macrophage turnover at inflammatory foci.
