Background
Rap1B is a small GTPase belonging to the Ras superfamily that functions as a critical molecular switch cycling between active GTP-bound and inactive GDP-bound states to regulate diverse cellular processes including cell adhesion, integrin activation, cell polarity, and platelet aggregation. Widely expressed across tissues, Rap1B plays a pivotal role in inside-out signaling by binding to effectors such as RIAM and RAPL to modulate integrin affinity for extracellular matrix ligands, thereby influencing cell migration, immune synapse formation, and hemostasis. Its activity is tightly controlled by guanine nucleotide exchange factors (GEFs) like CalDAG-GEFI and EPAC, as well as GTPase-activating proteins (GAPs) such as RAP1GAP and SPA-1, ensuring precise spatiotemporal regulation; dysregulation of Rap1B has been implicated in pathological conditions including bleeding disorders, cancer metastasis, and cardiovascular diseases, highlighting its importance as both a key physiological regulator and a potential therapeutic target.
