Background

RAD21 is one of the core subunits of the cohesin complex, encoded by the RAD21 gene on human chromosome 8, and is highly conserved in eukaryotes. As a key component of the cohesin trimer (SMC1A, SMC3, and RAD21), RAD21 interacts with SMC3 and SMC1A through its N-terminal and C-terminal domains, respectively, forming a ring-like structure that tightly entraps sister chromatids, ensuring accurate chromosome segregation during mitosis and meiosis. Beyond its classical cohesion function, RAD21 also plays a critical role in DNA damage repair. It undergoes ATR/ATM-mediated phosphorylation in response to DNA double-strand breaks, maintaining genomic stability by regulating homologous recombination and non-homologous end joining pathways. Additionally, RAD21 acts as a boundary factor guiding CTCF localization on the genome, participating in the delineation of topologically associating domains and the regulation of gene expression. In terms of disease, RAD21 gene mutations are a major cause of Cornelia de Lange syndrome (CdLS), which is characterized by multiple congenital malformations, craniofacial abnormalities, and intellectual disability. In cancer, RAD21 exhibits dual functions—its downregulation in certain leukemias and gliomas leads to insufficient cohesin function, promoting genome instability-driven carcinogenesis, whereas in other tumors, its overexpression enhances chemotherapy resistance by inhibiting apoptosis. Furthermore, RAD21 cleavage serves as a common switch for both apoptosis and separase-mediated sister chromatid separation. Under apoptotic stimulation, caspase-3 and granzyme B can directly cleave RAD21, accelerating nuclear disintegration. In summary, RAD21, as a multifunctional protein integrating chromosome cohesion, DNA repair, transcriptional regulation, and cell death, plays a central hub role in normal development and various disease processes.

Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.
