Flow cytometric analysis of Human PBMC labelling CLEC4A antibody at 1/200 (1 μg) dilution (Right panel) compared with a Rabbit IgG Isotype Control (Left panel). Goat Anti-Rabbit IgG DyLight Alexa Fluor® 488 was used as the secondary antibody. Then cells were stained with CD11b - Brilliant Violet 421™ antibody separately.

Background
CLEC4A (C-type lectin domain family 4 member A), also known as DCIR or CD367, is a type II transmembrane inhibitory receptor expressed mainly on dendritic cells and other myeloid cells; it contains an extracellular C-type lectin-like domain that binds glycan structures independent of calcium, a single transmembrane segment, and a cytoplasmic tail harboring an immunoreceptor tyrosine-based inhibitory motif (ITIM) which, upon ligand engagement, recruits phosphatases SHP-1/2 to down-regulate TLR signaling, suppress pro-inflammatory cytokine production, and maintain immune homeostasis, while also serving as an attachment factor for pathogens such as HIV-1.
