Background

Rab3D is a member of the small GTPase Rab family, belonging to the Rab3 subfamily, and is primarily localized on the membrane of secretory vesicles in secretory cells, playing a critical role in regulating vesicle trafficking and exocytosis. Through interactions with upstream regulators such as GEFs and GAPs, this protein cycles between the GTP-bound active state and the GDP-bound inactive state, thereby precisely controlling vesicle maturation, mobilization, docking, and fusion with target membranes. Rab3D is expressed in various tissues and is particularly abundant in pancreatic acinar cells and adipocytes, where its function is associated with the regulation of insulin-like growth factors and metabolic balance; it also participates in regulating neurotransmitter release, gastric acid secretion, and salivary secretion, making it an indispensable regulator of vesicle trafficking in exocrine and endocrine cells. Dysfunction or genetic deficiency of Rab3D is associated with multiple diseases; for example, Rab3D knockout mice exhibit increased bone mass and accelerated bone formation phenotypes, while downregulation of Rab3D expression in humans may be closely related to osteoporosis, obesity, and certain metabolic diseases, positioning it as an important molecular target for studying vesicle trafficking mechanisms and related metabolic disorders.
