Background

Rab27A is a highly specialized member of the Rab small GTPase family, serving as a key regulator of vesicle transport that is predominantly expressed in cells with secretory functions. Its core function is to act as a molecular switch that regulates the tethering, movement, and fusion of specific secretory vesicles with the plasma membrane. When bound to GTP, Rab27A recruits distinct effector proteins to regulate specific secretory processes in different cell types: in cytotoxic T cells and natural killer cells, it collaborates with effector molecules such as Munc13-4 and Slp2 to regulate the polarization and release of cytolytic granules toward the immune synapse, thereby playing a critical role in immune surveillance and clearance of virus-infected or tumor cells; in melanocytes, Rab27A forms a ternary complex with its effector proteins Melanophilin and Myosin Va, mediating the transport of melanosomes along the cytoskeleton toward the cell periphery, which is essential for normal pigmentation of skin and hair. Given its central role in secretory regulation, dysfunction of Rab27A is directly associated with various diseases: its gene mutations are the cause of Griscelli syndrome type 2, a rare autosomal recessive disorder characterized by partial albinism and immunodeficiency resulting from defective cytotoxic T lymphocyte function, which can be life-threatening in severe cases. Furthermore, aberrant expression of Rab27A has been observed in various tumors (such as breast cancer, melanoma, and pancreatic cancer), where it is closely associated with tumor invasion, metastasis, and immune evasion by influencing tumor cell secretory activity and tumor microenvironment remodeling, thus making it a potential intervention target in the field of tumor immunotherapy.

Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.
