Background
Lysine pyruvylation (Kpy) is a novel post-translational modification driven by the central glycolytic metabolite pyruvate, in which a pyruvoyl group is covalently attached to the ε-amino group of a lysine residue. This dynamic modification is regulated by the writer enzymes HAT1 and p300, and the eraser enzyme SIRT3. By coupling glycolytic flux to protein function, Kpy plays a pivotal role in epigenetic regulation and transcriptional activation, while also mediating metabolic-immune crosstalk, such as inhibiting type I interferon signaling under high-glucose conditions.
