Background

PTBP1 (Polypyrimidine Tract-Binding Protein 1, also known as hnRNP I) is a widely expressed RNA-binding protein belonging to the heterogeneous nuclear ribonucleoprotein family, playing critical regulatory roles in various RNA metabolic processes including pre-mRNA splicing, polyadenylation, mRNA stability, and translational regulation. The core structural feature of this protein is its four consecutive RNA recognition motifs (RRM1-4), among which RRM1 and RRM2 cooperatively bind to pyrimidine-rich RNA sequences (particularly polypyrimidine tracts) in an atypical manner, while RRM3 and RRM4 participate in auxiliary binding and protein-protein interactions. As a key regulator of alternative splicing, PTBP1 binds to polypyrimidine tracts flanking specific exons and inhibits spliceosome recognition of those exons through steric hindrance, thereby promoting exon skipping or inclusion. The most classic example occurs during neuronal differentiation, where the decline of PTBP1 leads to global switches in the splicing patterns of its target genes (such as NRCAM and REST), subsequently promoting neuronal maturation. In terms of subcellular localization, PTBP1 typically shuttles between the nucleus and cytoplasm, and its function exhibits high cell-type and developmental-stage specificity. Clinical studies have shown that PTBP1 is abnormally overexpressed in various malignant tumors (such as glioblastoma, hepatocellular carcinoma, pancreatic cancer, and breast cancer). By regulating the splicing of epithelial-mesenchymal transition-related genes, enhancing glycolytic metabolism, and inhibiting apoptosis, it promotes tumor cell proliferation, invasion, metastasis, and chemoresistance, making it an important candidate molecule for cancer diagnostic and prognostic markers as well as targeted therapy. Furthermore, dysregulation of PTBP1 is also closely associated with spinal muscular atrophy and certain viral infection processes, highlighting the central role of this protein in both physiological and pathological RNA processing networks.

Immunohistochemistry

Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.
