Background

PSMB5, also known as proteasome subunit beta type-5 or the X subunit of the 20S core particle, is a critical catalytic component of the 26S proteasome complex responsible for intracellular protein degradation via the ubiquitin-proteasome pathway. As one of the three distinct beta subunits possessing proteolytic activity within the 20S core, PSMB5 specifically exhibits chymotrypsin-like activity, cleaving peptide bonds adjacent to hydrophobic amino acid residues, which is essential for maintaining cellular homeostasis by removing damaged, misfolded, or regulatory proteins. This subunit plays a pivotal role in antigen processing for MHC class I presentation, thereby influencing immune surveillance, and serves as the primary molecular target for bortezomib and other proteasome inhibitor drugs used clinically to treat multiple myeloma and mantle cell lymphoma; furthermore, under inflammatory conditions induced by interferon-gamma, PSMB5 can be replaced by its inducible homolog LMP7 (PSMB8) to form the immunoproteasome, which alters cleavage specificity to optimize the generation of antigenic peptides for immune recognition.
