Background
PSD-95 (postsynaptic density protein 95) is a 95-kDa scaffolding protein enriched in the postsynaptic density of excitatory synapses, where its three N-terminal PDZ domains, an SH3 domain, and a C-terminal guanylate kinase-like domain assemble transmembrane receptors, ion channels, adhesion molecules, and downstream signaling enzymes into a dynamic macromolecular signaling lattice that couples synaptic activity to postsynaptic potentiation or depression and whose dysregulation contributes to autism, schizophrenia, and stroke.
