Background

PPP2R1A (Protein Phosphatase 2 Regulatory Subunit A Alpha) is a core scaffold subunit of the serine/threonine phosphatase PP2A, essential for assembling the catalytic C and regulatory B subunits into an active holoenzyme that dephosphorylates hundreds of substrates, thereby governing cell-cycle progression, apoptosis, metabolism, epithelial-mesenchymal transition, and antiviral signaling; it localizes to the cytoplasm and nucleus, is frequently mutated at hotspots in ovarian, endometrial and laryngeal cancers, undergoes post-translational succinylation that switches its function from tumor suppressor to oncogene by rewiring gluconeogenesis/lipogenesis, and its expression or mutation status modulates Wnt, TGF-β, PI3K-AKT and Hippo-YAP pathways, making it a key prognostic and therapeutic target in cancer and inflammatory diseases.

Immunohistochemistry

IHC shows positive staining in paraffin-embedded human testis. Anti-PPP2R1A antibody was used at 1/500 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.
