Background
POU2F3, also known as Oct-6 or Tst-1, is a transcription factor belonging to the class III POU domain family that plays a pivotal role in regulating gene expression during embryonic development and cellular differentiation, particularly within the nervous system and epidermal lineages. Characterized by its unique bipartite DNA-binding structure comprising a POU-specific domain and a POU homeodomain connected by a flexible linker, POU2F3 binds to specific octamer motifs in target gene promoters to modulate transcriptional activity, thereby driving critical processes such as Schwann cell myelination, neural crest formation, and the terminal differentiation of keratinocytes in the skin's suprabasal layers. Beyond its developmental functions, POU2F3 has garnered significant attention in oncology as a lineage-defining marker and potential therapeutic target in small cell lung cancer (SCLC) and other neuroendocrine tumors, where its aberrant overexpression contributes to tumor proliferation, survival, and the maintenance of a dedifferentiated state, making it a crucial molecule for understanding both normal tissue morphogenesis and malignant transformation mechanisms.

Immunohistochemistry
IHC shows positive staining in paraffin-embedded human gastric cancer. Anti-POU2F3 antibody was used at 1/125 dilution, followed by a HRP Polymer for Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.
