Background

PIN1 is a conserved 163-residue peptidyl-prolyl cis-trans isomerase composed of an N-terminal WW domain and a C-terminal catalytic PPIase domain, both recognizing pSer/Thr-Pro motifs; by selectively isomerizing these phosphorylated bonds it acts as a post-phosphorylation switch that reshapes the conformation, stability, localization and ubiquitination of hundreds of client proteins—ranging from p53 and cyclins to NF-κB and APP—thereby governing cell-cycle progression, transcription, immune signaling and neuronal homeostasis, while its frequent over-activation in cancer drives oncogenic pathways and its loss contributes to neurodegenerative diseases such as Alzheimer’s.

Immunocytochemistry

ICC shows positive staining in HEK293 cells. Anti- PIN1 antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 4% PFA and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
