Background
Phospho-TrkA (Tyr490) and phospho-TrkB (Tyr516) are the neurotrophin-activated, autophosphorylated forms of the receptor tyrosine kinases TrkA and TrkB; once these juxtamembrane tyrosines are phosphorylated they serve as docking platforms for adaptor proteins—TrkA-pTyr490 recruits FRS2 and Shc to trigger Ras/MAPK and PI3K/Akt cascades that mediate neuronal survival and differentiation, whereas TrkB-pTyr516 binds Shc to activate the same pathways plus PLCγ-driven PKC and Ca²⁺ signals that regulate synaptic plasticity—so antibodies detecting these phospho-sites are widely used as biomarkers for NGF- or BDNF-dependent Trk signaling in health and disease.
