Background

Phospho-TrkA (Tyr490) refers to the phosphorylation of tropomyosin receptor kinase A (TrkA) at tyrosine residue 490 (Y490), which is a key signal-initiating event following activation of TrkA by nerve growth factor (NGF). Upon phosphorylation at this site, it serves as a high-affinity docking site that specifically recruits the adapter protein Shc, thereby activating the downstream classical Ras-MAP kinase signaling cascade, playing a central role in regulating neuronal survival, differentiation, and synaptic plasticity. A 2013 study systematically confirmed, through the construction of mutant receptors, that Y490 is one of the two identified phosphorylated docking sites mediating TrkA signal transduction. Notably, phosphorylation at different sites mediates distinct signaling pathways—for example, phosphorylation at Y785 primarily activates the PLC-γ pathway, while phosphorylation at Y490 is critical for activating the PI3K pathway and for eliciting potent and sustained MAPK activation, the latter being essential for driving neuronal differentiation.
