Background

Phospho-SMAD2 (Ser465/467) refers to the activated form of the SMAD2 protein, in which the TGF-β type I receptor kinase (TβRI) specifically phosphorylates two serine residues at positions 465 and 467 within the C-terminal SSXS motif. This phosphorylation event is the critical step in canonical TGF-β signaling: it enables SMAD2 to dissociate from the activated receptor complex and form a heterodimeric complex with the co-mediator SMAD4, which then translocates into the nucleus to regulate target gene transcription. Notably, phosphorylation at both sites is required for this function, as mutation of either serine 465 or 467 prevents SMAD2 release from the receptor and blocks TGF-β-dependent signaling. Additionally, phosphorylation at Ser465 is prerequisite for phosphorylation at Ser467, and these C-terminal phosphoserines serve as a recognition site for SMAD4 binding.
