Background

Phospho-RIP3 (Thr231/Ser232) is the phosphorylated, catalytically active form of the 57 kDa serine/threonine kinase Receptor-Interacting Protein 3 (RIP3, RIPK3); dual phosphorylation at Thr231 and Ser232 within the conserved activation loop is induced by TNF-α, TRAIL or viral infection, converting RIP3 into a core driver of necroptosis—a programmed, caspase-independent inflammatory cell death pathway—via autophosphorylation and subsequent phosphorylation of downstream substrates such as MLKL, thereby assembling the necrosome complex and triggering membrane rupture, calcium influx and damage-associated molecular pattern (DAMP) release; detection of Phospho-RIP3 (Thr231/Ser232) by phospho-specific antibodies is therefore a key biomarker for necroptotic signaling in cancer, ischemia-reperfusion injury and inflammatory diseases.
