Background

Phospho-Rb (Ser780) refers to the specific modification form of the retinoblastoma protein (Rb) phosphorylated at its serine 780 residue. The Rb protein is a key regulator of the cell cycle G1/S checkpoint. In its unphosphorylated state, it halts cell cycle progression by binding to and inhibiting E2F transcription factors. When cells receive proliferative signals, the cyclin-dependent kinase 4/6-cyclin D complex specifically catalyzes the phosphorylation of the Rb protein at the Ser780 site. This event is an early and critical step in the inactivation process of the Rb protein. Phosphorylation at Ser780 directly disrupts the interaction between Rb and E2F, thereby relieving the inhibition. The released E2F then activates the transcription of a suite of genes necessary for S-phase entry, driving the cell cycle forward. Consequently, detecting the level of Phospho-Rb (Ser780) is commonly used to assess CDK4/6 activity and the state of cell proliferation, holding significant importance in cancer research and the development of targeted therapies, such as CDK4/6 inhibitors.
