Background

Phospho-PLK1 (Thr210) refers to the activated state of Polo-like kinase 1 following phosphorylation at the critical threonine 210 residue within its activation loop (T-loop), a modification widely recognized as the key molecular switch that drives PLK1 kinase activity. During cell cycle progression, Thr210 is primarily phosphorylated by Aurora A kinase and its cofactor Bora, a process further assisted by adaptor proteins such as WAC to facilitate proper mitotic entry. The phosphorylation level of Thr210 is tightly regulated: this modification is elevated during normal mitosis, but upon DNA damage, protein phosphatase 2A (PP2A) is recruited to remove the phosphate group from Thr210, thereby inhibiting PLK1 activity to arrest the cell cycle and initiate repair mechanisms. Functional studies have shown that the phosphomimetic T210D mutation, which results in sustained phosphorylation at this site, significantly enhances PLK1 kinase activity and leads to mitotic delay or arrest. Therefore, detecting the phosphorylation status of Thr210 (e.g., using the specific antibody #9062) is widely employed as a readout of PLK1 activity and holds significant value in cancer biology and cell cycle research.
