Background

Phospho-p70 S6 Kinase (Ser371) refers to the specifically activated form of p70 ribosomal protein S6 kinase (p70S6K, a key serine/threonine kinase) phosphorylated at its serine 371 residue (Ser371). As a central downstream effector of the mTORC1 (mechanistic target of rapamycin complex 1) signaling pathway, phosphorylation at this site is crucial for the full activation of p70S6K. Ser371 is located within the activation loop (T-loop) of the kinase domain. Its phosphorylation, typically catalyzed by mTORC1 itself or other upstream kinases, significantly enhances the kinase activity of p70S6K. The activated Phospho-p70 S6 Kinase (Ser371) then phosphorylates its downstream targets, such as ribosomal protein S6 and eIF4B, thereby directly promoting protein translation, ribosome biogenesis, and cell growth. Consequently, detecting the phosphorylation level at Ser371 is a commonly used biomarker for assessing mTORC1 pathway activity and studying cell proliferation, metabolic regulation, as well as drug target responses in related diseases such as cancer and metabolic disorders.
