Background
Phospho-Myosin Light Chain 2 (Ser19) refers to the specific post-translational modification of the regulatory myosin light chain (MLC2, also known as MLC20 in smooth muscle or MLC2a/2b in striated muscle) where a phosphate group is added to the serine residue at position 19. This phosphorylation event is primarily catalyzed by Myosin Light Chain Kinase (MLCK) in response to increased intracellular calcium levels, although Rho-associated kinase (ROCK) can also contribute, particularly in smooth muscle tone regulation. The addition of the phosphate group at Ser19 induces a conformational change in the myosin head, enabling it to interact with actin filaments and initiate ATP-dependent cross-bridge cycling, which is the fundamental mechanism driving muscle contraction and non-muscle cell motility, such as cytokinesis and cell migration. Consequently, detection of Phospho-MLC2 (Ser19) is widely used as a key biomarker for assessing cellular contractility and cytoskeletal dynamics in various physiological and pathological contexts.
