Background
Phospho-MLKL (Ser345) is the activated, phosphorylated form of the pseudokinase mixed-lineage kinase domain-like protein (MLKL) at serine-345, a critical post-translational modification catalyzed by the necroptotic kinase RIPK3 that triggers MLKL’s oligomerization, translocation to plasma membrane phosphoinositides, and subsequent membrane disruption, thereby executing the terminal, pro-inflammatory cell death pathway known as necroptosis; detection of phospho-MLKL(Ser345) by phospho-specific antibodies thus serves as a specific biomarker for ongoing necroptotic signaling in experimental models and human pathologies including ischemia–reperfusion injury, inflammatory diseases, and certain cancers.
