Background

Phospho-IKKα/β (Ser176/180) refers to the phosphorylated, catalytically active forms of the IκB kinase (IKK) catalytic subunits IKKα (at Ser176/Ser180) and IKKβ (at Ser177/Ser181); these activating phosphorylation events, induced by inflammatory cytokines such as TNF-α and IL-1β, trigger a conformational switch that fully activates the IKK complex, enabling it to phosphorylate the NF-κB inhibitor IκBα and thus liberate the transcription factor NF-κB for nuclear import and pro-inflammatory gene expression.

Immunohistochemistry

IHC shows positive staining in paraffin-embedded human breast cancer. Anti- Phospho-IKKα/β (Ser176/180) antibody was used at 1/500 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.
