Background
Phospho-Erk5 (Thr218/Tyr220) is a phosphorylated form of the extracellular signal-regulated kinase 5 (Erk5), also known as big MAP kinase 1 (BMK1). It is activated through dual phosphorylation at threonine 218 and tyrosine 220 within its TEY activation motif by upstream MAP kinase kinases MEK5. This phosphorylation event is essential for Erk5 catalytic activity and its subsequent nuclear translocation, where it regulates gene expression involved in cell proliferation, survival, and differentiation. Phospho-Erk5 serves as a specific biomarker for Erk5 pathway activation in various cellular processes and disease states, including cancer progression and cardiovascular development.
