Background

Phospho-Cdc25A (Ser124) refers to the modified form of the cell cycle regulatory protein CDC25A following phosphorylation at serine residue 124. CDC25A is a dual-specificity protein phosphatase that primarily drives cells into S phase and promotes DNA replication during the G1/S transition by removing inhibitory phosphate groups from cyclin-dependent kinases such as CDK2. The Ser124 site can be phosphorylated by the checkpoint kinases CHK1 and CHK2 within DNA damage and stress response pathways, as well as by p38 MAPK under conditions such as osmotic stress. Phosphorylation at this site is generally associated with the ubiquitination and proteasomal degradation of CDC25A, leading to reduced protein levels, suppression of CDK2 activity, and cell cycle checkpoint arrest at the G1/S or S phase, thereby preventing damaged cells from continuing to divide. Consequently, Phospho-Cdc25A (Ser124) is commonly used as a marker for stress-induced phosphorylation of CDC25A and checkpoint activation, and is employed in Western blotting, immunohistochemistry, and immunoprecipitation assays to assess DNA damage, replication stress, or the status of tumor-associated signaling pathways.

Immunohistochemistry

IHC shows positive staining in paraffin-embedded human testis. Anti-Phospho-Cdc25A (Ser124) antibody was used at 1/400 dilution, followed by a HRP Polymer for Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.
