Background
Phospho-c-Kit (Tyr721) refers to the phosphorylated form of the c-Kit receptor tyrosine kinase at tyrosine residue 721 within its intracellular juxtamembrane domain. Upon binding of its ligand stem cell factor (SCF), c-Kit undergoes autophosphorylation, including at Tyr721, which serves as a critical docking site for the p85 regulatory subunit of phosphatidylinositol 3-kinase (PI3K). This phosphorylation event initiates downstream PI3K/Akt signaling cascades, thereby regulating essential cellular processes such as survival, proliferation, differentiation, and migration in hematopoietic stem cells, melanocytes, mast cells, and germ cells. Aberrant phosphorylation at this site has been implicated in various pathological conditions, including certain gastrointestinal stromal tumors (GISTs) and myeloproliferative disorders.
