Background

Phospho-alpha-Adducin (Ser12) refers to a specific modified form of α-adducin in which the serine residue at position 12 (Ser12) undergoes phosphorylation. α-Adducin is a cytoskeleton-associated protein that is typically involved in processes such as capping the barbed ends of actin filaments, promoting the binding of spectrin to actin, and synaptic assembly. Phosphorylation at the Ser12 site is specifically catalyzed by cyclin-dependent kinase 1 (CDK1) during mitosis and, together with phosphorylation at another site (Ser355), enables α-adducin to bind to the unconventional myosin Myosin-X (Myo10) and subsequently localize to the mitotic spindle. This phosphorylation event is critical for proper spindle assembly and chromosome alignment, and its disruption leads to spindle malformation, elongation, or multipolarity, resulting in chromosome misalignment. Therefore, Phospho-alpha-Adducin (Ser12) not only serves as an important marker for cells entering mitosis but also reflects the key role of α-adducin in regulating cell proliferation and division.
