Background

The Phospho-Akt Substrate (RXRXXS/T) motif is the core sequence specifically recognized and phosphorylated by protein kinase B (Akt). In this motif, R represents arginine, X represents any amino acid, and S/T represents the phosphorylation site serine or threonine. The characteristic arginine residues (RXR) are responsible for specific binding to the catalytic domain of Akt, while the C-terminal serine or threonine (S/T) is the site where Akt catalyzes the phosphorylation reaction. This highly conserved motif is widely found in various downstream substrate proteins of Akt, such as GSK-3β and FOXO transcription factors, and serves as a central hub regulating key signaling pathways involved in cell survival, proliferation, and metabolism. Detecting the phosphorylation level of this motif directly reflects the in vivo activity status of Akt kinase.
