IHC shows positive staining in paraffin-embedded mouse spleen. Anti-PF4 antibody was used at 1/2000 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

Background
Platelet factor 4 (PF4) is a 70–amino-acid, 7.8-kDa CXC chemokine stored in platelet α-granules and released upon activation, where it binds glycosaminoglycans such as heparin with ultra-high affinity, neutralizes the anticoagulant activity of heparin by bridging and clustering it on platelet or endothelial surfaces, and forms stable tetramers that can subsequently assemble into ultra-large immune complexes recognized by anti-PF4/heparin antibodies in heparin-induced thrombocytopenia (HIT), while also exerting broader immune-modulatory effects by recruiting and activating leukocytes, promoting neutrophil extracellular trap (NET) formation, enhancing monocyte adhesion and phagocytosis, and amplifying thrombo-inflammation in conditions such as sepsis, COVID-19, and atherosclerosis through interaction with receptors including CXCR3, TLR4, and FcγRIIA.

ICC shows positive staining in mouse splenocytes cells. Anti-PF4 antibody was used at 1/100 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 4% PFA and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
